Les premières mentions d'une cathédrale remontent au 12e siècle, lorsqu'il existait une église dédiée à Saint-Pierre l'Apôtre. Le bâtiment n'étant pas assez grand pour accueillir un grand nombre de fidèles, il a été démoli un siècle après sa construction pour faire place à un nouveau bâtiment, qui a duré jusqu'à l'ère napoléonienne. Le nouveau bâtiment était dédié à Saint-Marc et régi par l'ordre dominicain.
En 1797, l'église fut confisquée par Napoléon Bonaparte et ses troupes, qui en firent leur quartier général ; il devint rapidement nécessaire de démolir la structure pour des raisons militaires. Les travaux de reconstruction durent de 1807 à 1810 et nécessitent d'importantes interventions de l'architecte Cristoforo Vallizzone. Le nouveau bâtiment avait complètement perdu ses connotations gothiques d'origine, prenant une apparence néoclassique. Lors de sa réouverture en 1810, la cathédrale d'Alexandrie a été confirmée comme étant la cathédrale de Saint-Pierre et de Saint-Marc. La façade de l'édifice sacré n'a été achevée qu'en 1820 ; les fresques qui symbolisent la vie de l'apôtre saint Pierre sont intéressantes. La décoration intérieure du bâtiment a été réalisée dans les années 1920. Il y a plusieurs œuvres d'intérêt à l'intérieur du bâtiment.
